利用TCO和Tz之間快速的正交切割反應,把Tz搭載于腫瘤靶向性的納米材料單壁碳納米管SWCNTs上(Tz@SWCNTs),實現對腫瘤的預靶向激活。其主要原理:利用通透性增強與滯留效應,使Tz@SWCNTs在腫瘤組織中積累。通過TCO前藥分子或探針,由在腫瘤處富集的Tz進行正交切割。
通過研究tAMC與Tz@SWCNTs之間的釋放動力學,相比于mTZ小分子,Tz@SWCNTs對tAMC的衰減反應性更強,且能在生理環境下,對MCF-7癌細胞進行靶向并激活tAMC熒光。
將陽離子染料半菁HCA和TCO相連得到近紅外熒光探針tHCA用于通過生物正交釋放反應的激活近紅外熒光探針。該探針在生理環境下因Tz@SWCNTs的激發快速釋放,并在720nm下發出強熒光。
